چکیده

Abstrad It has been proposed that the transfer of growth regulatory signals via gap jundions is important in the control of proliferation. In confluent 1 01#{189} cells, growth control is enhanced by retinoids; this adion is highly correlated with up-regulated gap jundional communication (GJC). Treatment of quiescent 1 0T#{189} cells with transforming growth fador (TGF) flu and TGF-fi2 resulted in elevated levels of proliferation together with increased expression of connexin43 protein and elevated GJC. Connexin43 was localized into plaques in regions of cell-cell contad; such plaques were found at high frequency in treated cells but only rarely in control cultures. These data illustrate that increased cell proliferation can occur in the presence of enhanced GJC, a result at variance with our previous results with retinoids. We suggest that either the proliferative stimulus of TGF-1J is sufficient to overwhelm any antiproliferative effed of GJC or that under conditions of TGF-fi stimulation, jundions convey net proliferative stimuli.

Transforming growth factor-beta 1 and forskolin modulate gap junctional communication and cellular phenotype of cultured Schwann cells.

Following peripheral nerve injury, Schwann cells undergo a series of cellular alterations that are thought to assist the regenerative process. Some of these changes are stimulated by the local release of cytokines and mitogenic factors. To test the hypothesis that cytokine regulation of gap junctional communication between cells helps to coordinate Schwann cell responses, cultured rat Schwann c...

Hormonal regulation of connexin 43 expression and gap junctional communication in human osteoblastic cells.

We have recently shown that connexin 43 (Cx43), a major gap junction protein in osteoblasts, is expressed with an increase in cell density (CHIBA, H. et al. (1993). Cell Struc. Funct., 18: 419-426). In the present study, we examined what kinds of hormones and cytokines regulate the gap junction protein in osteoblastic cells, using a human osteoblastic cell line (SV-HFO) after reaching a conflue...

Metalloproteinases, Mechanical Factors and Vascular Remodeling

Chronic increases in arterial blood flow elicit an adaptive response of the arterial wall, leading to vessel enlargement and reduction in wall shear stress to physiological baseline value. Release of nitric oxide from endothelial cells exposed to excessive shear is a fundamental step in the remodeling process, and potentially triggers a cascade of events, including growth factor induction and m...

Human Leukocyte Antigen-G Expression on Dendritic Cells Induced by Transforming Growth Factor-β1 and CD4+ T Cells Proliferation

Background: During antigen capture and processing, mature dendritic cells (DC) express large amounts of peptide-MHC complexes and accessory molecules on their surface. DC are antigen-presenting cells that have an important role in tolerance and autoimmunity. The transforming growth factor-beta1 (TGF-β1) cytokine has a regulatory role on the immune and non-immune cells. The aim of this study is ...

Differential effects of transforming growth factor beta 1 on the growth of poorly and highly metastatic murine melanoma cells.

We have examined the effects of transforming growth factor beta 1 (TGF-beta 1) on the growth of paired murine melanoma cell clones that differ with respect to their experimental metastatic potential. Neither poorly (clone 16) nor highly (clone M2) metastatic cells were capable of anchorage-independent growth in 0.3% agar/Dulbecco's modified Eagle's medium in the absence of serum. However, both ...
